Abstract
(EN) The present invention relates to compositions and methods for regulating cellular process, such as the cell-cycle, cell proliferation, cell adhesion, and cancer, especially processes that involve TGF$g(b) signalling. In preferred embodiments, the invention relates to methods of identifying agents comprising one or more of the following steps, a) administering a test agent to cells, wherein the cells have reduced or no TGF$g(b)/SMAD activity, b) administering to said cells an amount of TGF$g(b), or an amount of a ligand which acts as an agonist or partial agonist on the TGF$g(b) receptor, effective to produce a phenotype; and c) determining whether TGF$g(b) elicits the phenotype in the presence of the test agent, whereby said agent is identified as regulating said phenotype of the cells.
(FR) La présente invention concerne des compositions et des procédés permettant de réguler des processus cellulaires tels que le cycle cellulaire, la prolifération cellulaire, l'adhésion cellulaire et le cancer, en particulier des processus dans lesquels est impliquée la signalisation TGF-bêta (facteur de croissance transformant bêta). Dans des modes de réalisation préférés, l'invention se rapporte à des procédés d'identification d'agents, selon lesquels : a) on administre un agent d'essai à des cellules présentant une activité TGF-bêta/SMAD réduite ou nulle ; b) on administre aux cellules précitées une quantité efficace pour produire un phénotype de TGF-bêta ou d'un ligand agissant comme agoniste ou agoniste partiel sur le récepteur TGF-bêta; et c) on détermine si le TGF-bêta élicite le phénotype en présence de l'agent d'essai, l'agent précité étant identifié, si tel est le cas, comme régulant ce phénotype cellulaire.
