Abstract:
(EN) The invention relates to a method for the production of proteinogenic or non-proteinogenic L-amino acids, especially L-phosphinothricin, from their racemic N-acetyl-D,L-derivatives. The method is characterized in that: (a) N-acetyl-L-derivatives of the corresponding L-amino acids are selectively deacetylated by enzymatic racemate cleavage by means of isolated recombinant enzymes while the N-acetyl-D-derivatives of the corresponding D-amino acids are not deacetylated and (b) the deacetylated L-amino acids are preparatively separated from the non-deacetylated N-acetyl-D-derivatives and/or the incompletely deacetylated N-acetyl-L-derivatives.
(FR) L'invention concerne un procéde de fabrication d'acides aminés L protéinogènes ou non-protéinogènes, en particulier de phosphinothricine L, à partir de leurs dérivés-D,L-N-acétyliques racémiques. Ce procédé est caractérisé en ce que ; (a) des dérivés-L-N-acétyliques des acides aminés L correspondants sont déacétylés sélectivement par séparation racémique enzymatique au moyen d'enzymes recombinantes isolées, alors que des dérivés-D-N-acétylés des acides aminés D correspondants ne sont pas déacétylés ; et (b) les acides aminés L déacétylés obtenus sont séparés préparativement des dérivés-D-N-acétyliques non déacétylés et des dérivés-L-N-acétyliques partiellement déacétylés.
(DE) Ein Verfahren zur Herstellung von proteinogenen oder nicht-proteinogenen L-Aminosäuren, insbesondere L-Phosphinothricin, aus deren racemischen N-Acetyl-D,L-Derivaten ist dadurch gekennzeichnet, daß (a) durch eine enzymatische Racemat-Spaltung mittels isolierter, rekombinanter Enzyme selektiv N-Acetyl-L-Derivate der korrespondierenden L-Aminosäuren deacetyliert werden, während N-Acetyl-D-Derivate der korrespondierenden D-Aminosäuren nicht deacetyliert werden und (b) die erhaltenen deacetylierten L-Aminosäuren präparativ von den nicht deacetylierten N-Acetyl-D-Derivaten und/oder den nicht vollständig deacetylierten N-Acetyl-L-Derivaten getrennt werden.
