The invention provides modified reverse transcriptase polypeptides (Types I, II, and III), along with polynucleotides encoding such polypeptides, vectors containing such polynucleotides and host cells transformed with those polynucleotides. The modified RTs typically exhibit improved stability and/or improved solubility, relative to naturally occurring reverse transcriptases. The modified RTs are also found in a variety of forms, such as monomers as well as both homo- and hetero-multimers. The modified RTs may be used in any one or more of the methods known to benefit from reverse transcriptase activity, such as cDNA synthesis, and amplification techniques such as PCR and RAMP.

La présente invention concerne des polypeptides de types I, II et III, en l'occurrence des transcriptases inverses modifiées ainsi que des polynucléotides codant de tels polypeptides, des vecteurs contenant de tels polynucléotides et des cellules hôtes transformées avec ces polynucléotides. Ces transcriptases inverses modifiées font preuves de qualités améliorées de stabilité et/ou de solubilité par rapport aux transcriptases inverses existant naturellement. Ces transcriptases inverses modifiées se trouvent également sous diverses formes, et notamment des monomères ainsi que des homomultimères et des hétéromultimères. Les transcriptases inverses modifiées de l'invention convient pour l'un ou l'autre des procédés connus exploitant l'activité transcriptase inverse, et notamment la synthèse d'ADN complémentaire, et des techniques d'amplification et notamment PCR et RAMP.
