The present invention relates to rapid, quantitative, specific, high through-put methods for screening test substances for their ability to inhibit activity of an ouabain-resistant Na+-K+-ATPase involved in a variety of biological processes such as regulation of osmotic balance and cell volume, maintenance of the resting membrane potential, establishment of the ionic composition of cerebrospinal fluid and aqueous humor, electrical activity of muslce and nerve, and receptor-mediated endocytosis, cardiac muscle contractility, neurotransmitter metabolism and vascular muscle cell contraction. These methods can be employed to identify compounds for use in therapeutic applications for disease processes in which dysfunction of the Na+-K+-ATPase contributes to a pathological process. The present invention also includes kits which are used in the methods provided herein. The present invention further includes methods of treating or preventing diseases or disorders which are associated with dysfunction of the Na+-K+-ATPase.

La présente invention concerne des procédés rapides, quantitatifs, spécifiques, et à grande échelle, permettant de rechercher dans des substances d'essai leur capacité d'inhibition de l'activité d'une Na+-K+-ATPase résistant à la ouabaïne. Celle-ci participe à une variété de processus biologiques tels que la régulation de l'équilibre osmotique et du volume cellulaire, le maintien du potentiel de repos de la membrane, l'établissement de la composition ionique du liquide céphalo-rachidien et de l'humeur aqueuse, l'activité électrique des muscles et des nerfs, l'endocytose induite par des récepteurs spécifiques, la contractilité des muscles cardiaques, le métabolisme neurotransmetteur et la contraction cellulaire des muscles vasculaires. Ces procédés permettent d'identifier des composés s'utilisant dans des applications thérapeutiques du processus morbide dans lequel le dysfonctionnement de la Na+-K+-ATPase contribue à un processus pathologique. La présente invention comprend également des kits qui s'utilisent dans les procédés décrits, et concerne en outre des procédés de traitement ou de prévention des maladies ou des troubles associés au dysfonctionnement de la Na+-K+-ATPase.
