(EN)The present invention provides a method for promoting the survival and/or expansion of hematopoietic stem cells in culture, comprising culturing the cells in a culture including an effective amount of a mpl ligand, (particularly thrombopoietin), an flt3 ligand and interleukin 6 (IL6). Also provided is a method for modifying hematopoietic stem cells comprising contacting a gene delivery vehicle comprising a polynucleotide sequence with a population of hematopoietic stem cells cultured in the presence of an effective amount of thrombopoietin and an flt3 ligand. Additionally methods of restoring hematopoietic capability in a subject are claimed.
(FR)La présente invention concerne une méthode favorisant la survie et/ou la multiplication de cellules souches hématopoïétiques en culture. Ladite méthode consiste à cultiver les cellules dans un milieu de culture comprenant une quantité efficace d'un ligand mpl, (notamment de thrombopoïétine), d'un ligand ftl3 et d'interleukine 6 (IL6). L'invention concerne également une méthode de modification des cellules souches hématopoïétiques, consistant à mettre un vecteur d'apport de gènes renfermant une séquence polynucléotidique au contact d'une population de cellules souches hématopoïétiques cultivées en présence d'une quantité efficace de thrombopoïétine et d'un ligand ftl3. L'invention concerne enfin des méthodes permettant de restaurer la capacité hématopoïétique d'un sujet.
