Abstract
(EN) A process and apparatus in which cryoprotectant in cryopreserved samples is removed with the use of turbulence and convective-dispersion around the cells within the primary cryopreservation container are described. Convective-dispersion is applied via the combined action of gravity and pulsative transmembrane flow of buffer through the pores of containers fabricated from appropriate membrane material. Use of mechanical forces to aid the process of cryoprotectant removal permits the benefits of slow cryoprotectant removal to be retained, while the actual removal time is decreased.
(FR) L'invention concerne un procédé et un appareil permettant d'extraire un cryoprotecteur d'échantillons cryoconservés. Selon ledit procédé, on extrait le cryoprotecteur en utilisant la turbulence et la dispersion convective se produisant autour des cellules, à l'intérieur du récipient primaire de cryoconservation. La dispersion convective est produite par l'action combinée de la gravité et de l'écoulement pulsatoire transmembranaire de tampon à travers les pores de récipients fabriqués à partir d'un matériau pour membranes approprié. L'utilisation de forces mécaniques pour assister le processus de l'extraction du cryoprotecteur permet de conserver les avantages d'une extraction lente de cryoprotecteur tout en ayant un temps d'extraction réel nettement réduit.
