Abstract:
(EN) Compositions and methods are provided for determining the presence of an antibiotic resistant $i(mecA) gene in a biological sample, comprising the general steps of (a) treating cells contained within a biological sample to expose target single-stranded nucleic acid molecules; (b) reacting the target single-stranded nucleic acids with a scissile link-containing nucleic acid which is complementary to a portion of an antibiotic resistant $i(mecA) gene, and with an enzyme which cleaves double-stranded target-probe complexes, under conditions which allow the target and probe to hybridize to each other to form a double-stranded target-probe complex, the enzyme molecule being capable of cleaving the scissile link of the target-probe complex such that one or more fragments of the nucleic acid probe released from said complex; and (c) determining whether cleaved portions of the detecting probe fragments released from said nucleic acid probe are produced, and thereby detecting the presence of the antibiotic resistant $i(mecA) gene.
(FR) L'invention concerne des compositions et des procédés pour déterminer la présence d'un gène $i(mecA) résistant aux antibiotiques dans un échantillon biologique. Ces procédés consistent à (a) traiter les cellules contenues dans un échantillon biologique pour exposer les molécules d'acide nucléique cibles à simple brin; (b) faire réagir les acides nucléiques cibles à simple brin avec un acide nucléique contenant une liaison pouvant être scindée qui est complémentaire d'une partie d'un gène $i(mecA9) résistant aux antibiotiques, et avec un enzyme qui clive les complexes cible-sonde à deux brins, dans des conditions qui permettent à la cible et à la sonde de s'hybrider l'une à l'autre pour former un complexe cible-sonde à deux brins, la molécule enzymatique pouvant assurer le clivage de la liaison pouvant être scindée du complexe cible-sonde, de telle sorte que un ou plusieurs fragments de la sonde d'acide nucléique sont libérés de ce complexe, et (c) déterminer si les parties clivées de la sonde d'acide nucléique sont produites et ainsi détecter la présence d'un gène $i(mecA) résistant aux antibiotiques.
