Abstract:
(EN) Impaired TPO-mediated platelet protein tyrosine phosphorylation was consistently observed in patients with polycythemia vera (PV) as well as those with idiopathic myelofibrosis (IMF). Impaired TPO-induced protein tyrosine phosphorylation in PV and IMF platelets was uniformly associated with markedly reduced or absent expression of the extracellular domain of Mpl detectable by the use of an antibody. Thus the reduced detectability of Mpl can be used as a marker of PV and IMF. The abnormality appears to distinguish PV from other forms of erythrocytosis and may be involved in the platelet function defect associated with PV.
(FR) On a pu observer de façon régulière des défauts de phosphorylation de la tyrosine protéique des plaquettes, induits par le TPO, chez des patients atteints de polycythemia vera (PV) et chez ceux atteints de myélofibrose idiopathique (IMF). Les défauts de phosphorylation de la tyrosine protéique des plaquettes PV et IMF, induits par le TPO était uniformément associée à une expression notablement réduite ou absente du domaine cellulaire du Mpl. De ce fait, la moindre détectabilité du Mpl peut servir de marqueur du PV et de l'IMF. Cette anomalie, qui apparaît comme une possibilité de distinguer le PV d'autres formes d'érythrocytose, peut jouer un rôle dans les défaut de fonctionnement des plaquettes liés au PV.
