Abstract:
(EN) The present invention refers to a method that allows the use of filamentous bacteriophages to detect the presence of molecules of interest in biological samples. It consists of a series of combined operations, which are drawn up and defined according to the characteristics of the phages employed. These have, exposed on the surface of the capsid, at least one molecule, generally but not exclusively of proteic nature, capable of binding at least one molecule of interest present in said biological sample. Detection of the presence of said molecule takes place, according to the method of the invention, using the association between the molecule exposed on the surface of the phage and the genome of the phage itself. The ability to form specific complexes typical of the molecule exposed on the capsid, does enable formation of phage-molecule of interest complexes, which can be detected by means of amplification of known sequences in the phage DNA. The method according to the invention is extremely sensitive and can be used in particular for diagnostic and prognostic purposes.
(FR) L'invention concerne un procédé permettant l'utilisation de bactériophages filamenteux pour la détection de la présence de molécules à analyser dans des échantillons biologiques. Ledit procédé consiste en une série d'opérations combinées qui sont décrites et définies en fonction des caractéristiques des phages employés. Lesdits phages comprennent, lorsqu'ils sont exposés sur la surface du capside, au moins une molécule, généralement, mais pas exclusivement, de nature protéique, capable de fixer au moins une molécule à analyser présente dans ledit échantillon biologique. La détection de la présence de ladite molécule s'effectue, selon le procédé de l'invention, au moyen de l'association entre les molécules exposées à la surface du phage et du génome du phage lui-même. La capacité de former des complexes spécifiques typiques de la molécule exposée sur le capside, permet la formation de molécule-phage constituée des complexes à analyser, qui peut être détectée par l'amplification des séquences connues dans l'ADN du phage. Le procédé de l'invention est extrêmement sensible et peut être utilisé notamment à des fins diagnostiques et pronostiques.
