Abstract:
(EN) The invention concerns a method for demonstrating an alteration in a DNA sequence, characterised in that: a) said DNA sequence is brought in the presence of a composition comprising at least a compound identifying the type of alteration concerned, called ligand, in a medium ensuring the identification; b) revealing the identification of the alteration by said ligand. The invention is useful for demonstrating the genotoxic properties of a medium by measuring the alteration produced by the medium on the DNA sample, and for demonstrating an alteration of the DNA repair system.
(FR) La présente invention concerne un procédé de mise en évidence d'une altération d'une séquence d'ADN, caractérisé en ce que: a) on met ladite séquence d'ADN en présence d'une composition comportant au moins un composé reconnaissant le type d'altération en cause, dénommé ligand, dans un milieu assurant la reconnaissance, b) on révèle la reconnaissance de l'altération par ledit ligand. Application à la mise en évidence des propriétés génotoxiques d'un environnement par mesure de l'altération que l'environnement produit sur l'ADN de l'échantillon, ainsi que la mise en évidence d'une altération du système de réparation de l'ADN.
