Abstract:
(EN) The invention provides a method for preparing a nucleic acid binding protein of the Cys2-His2 zinc finger class capable of binding to a nucleic acid quadruplet in a target nucleic acid sequence, wherein binding to base 4 of the quadruplet by an $g(a)-helical zinc finger nucleic acid binding motif in the protein is determined as follows: if base 4 in the quadruplet is A, then position +6 in the $g(a)-helix is Gln and position ++2 is not Asp; and if base 4 in the quadruplet is C, then position +6 in the $g(a)-helix may be any residue, as long as position ++2 in the $g(a)-helix is not Asp.
(FR) L'invention concerne une méthode de préparation d'une protéine de liaison d'acide nucléique de la classe de doigts à zinc Cys2-His2 capable de se lier à un quadruplet d'acide nucléique dans une séquence d'acide nucléique cible, la liaison à la base 4 du quadruplet par un motif de liaison d'acide nucléique de doigt à zinc $g(a)-hélicoïdal étant déterminée comme suit: si la base 4 dans le quadruplet est A, alors la position +6 dans l'hélice $g(a) est Gln et la position ++2 n'est pas Asp; et si la base 4 dans le quadruplet est C, alors la position +6 dans l'hélice $g(a) peut être n'importe quel résidu, tant que la position ++2 dans l'hélice $g(a) n'est pas Asp.
