Abstract:
(EN) Ligand-aminodextran-(phycobiliprotein or tandem dye) conjugates useful for detection of a desired target biological material by providing an enhanced fluorescent signal are described. Also described is a method for a single-measurement quantification of multiple populations of cells based upon the labeling of different pairs of cell populations, each pair containing mutually exclusive cell receptors which are expressed at substantially similar receptor densities with labeled ligands for each receptor. One cell population is labeled with a ligand capable of binding to a first cell surface receptor which ligand is directly conjugated to a fluorescent phycobiliprotein or tandem dye; and a second cell population is labeled with a ligand capable of binding to a second cell surface receptor, which ligand is cross-linked by an aminodextran to a fluorescent phycobiliprotein or tandem dye.
(FR) L'invention concerne des conjugués de ligand-dextrane aminé-(phycobiliprotéine ou colorant en tandem) s'utilisant dans la détection d'une matière biologique cible voulue, par émission d'un signal fluorescent amélioré. L'invention concerne également un procédé de quantification par mesure unique de plusieurs populations de cellules sur la base du marquage de différentes paires de populations de cellules, chaque paire contenant des récepteurs de cellules mutuellement exclusifs qui sont exprimés à des densités de récepteur sensiblement similaires à celles de ligands marqués pour chaque récepteur. Une population de cellules est marquée avec un ligand capable de se lier à un premier récepteur de surface de cellule, ce ligand étant directement conjugué à une phycobiliprotéine fluorescente ou à un colorant en tandem; et une deuxième population de cellules est marquée avec un ligand capable de se lier à un deuxième récepteur de surface de cellule, ce ligand étant réticulé par un dextrane aminé à une phycobiliprotéine fluorescente ou à un colorant en tandem.
