Abstract:
(EN) A method and apparatus are provided for differentiating cells in an automated particle analyzer (10), such as for use in hematological analysis, by measuring apparent cell size (or volume), membrane integrity and intracellular complexity ('voltaic fragility'). The cells are suspended in a selected reagent mixture. A predetermined dc voltage is applied across a sensing orifice (36) to thereby create a dc electric field within and around the orifice (36), and the reagent mixture is passed through the center of the orifice (36). The magnitude of the change in current flowing through dc electric field is measured therethrough, and based on the measured change, a signal is generated for each cell which is indicative of the apparent complexity of the cell. A visual display (54) is generated based on the signals facilitating differentiation of the cells.
(FR) On décrit un procédé et un appareil permettant de différencier des cellules dans un analyseur automatique (10) de particules, tel que celui qu'on utilise dans l'analyse hématologique, le procédé comprenant la mesure de la taille apparente (ou du volume) des cellules, de l'intégrité des membranes et de la complexité intracellulaire ('fragilité voltaïque'). Les cellules se trouvent en suspension dans un mélange réactif sélectionné. On applique une tension en courant continu prédéterminée au niveau d'un orifice (36) de détection pour créer ainsi un champ électrique à courant continu à l'intérieur et autour de l'orifice (36) puis on fait passer le mélange réactif par le centre de l'orifice (36). L'importance du changement se produisant dans le courant s'écoulant à travers le champ électrique à courant continu est mesurée dans ce dernier et en fonction du changement mesuré pour chaque cellule est généré un signal qui indique la complexité apparente de la cellule. Un affichage visuel (54) est généré sur la base des signaux permettant d'effectuer la différenciation des cellules.
