Abstract:
(EN) A recombinant lentiviral vector expression system comprises a first vector that comprises a nucleic acid sequence of at least part of the Equine Infectious Anemia Virus (EIAV) genome. The vector contains at least one defect in at least one gene encoding the EIAV structural protein, but is preferably a $i(gag/pol) expression vector. The expression system further comprises a second vector, also comprising a nucleic acid sequence of at least part of the Equine Infectious Anemia Virus (EIAV) genome, and additionally containing a multiple cloning site wherein a heterologous gene may be inserted. The expression system also comprises a third vector which expresses a viral envelope protein. The first and third vector are packaging signal-defective. When the expression system is transfected into a lentivirus-permissive cell, replication-defective EIAV particles may be produced, which particles are useful in delivering heterologous genes to a broad range of both dividing and non-dividing cells.
(FR) La présente invention concerne un système d'expression de vecteur lentiviral recombinant comprenant un premier vecteur comportant une séquence d'acide nucléique d'au moins une partie du génome du virus d'anémie infectieuse des équidés (EIAV). Ce vecteur contient au moins une anomalie dans au moins un gène codant une protéine structurale EIAV mais est, de préférence, un vecteur d'expression $i(gag/pol.) Le système d'expression comprend, en outre,$i( )un second vecteur également comportant une séquence d'acide nucléique d'au moins une partie du génome du virus d'anémie infectieuse des équidés (EIAV) ainsi qu'un site de clonage multiple dans lequel on peut insérer un gène hétérologue. Le système d'expression comprend également un troisième vecteur qui exprime une protéine d'enveloppe virale. Les premier et second vecteurs sont déficients en signaux d'encapsulation. Lorsqu'on transfecte le système d'expression dans une cellule permettant la multiplication d'un lentivirus, on peut produire des particules EIAV inaptes à la réplication, lesquelles sont utilisées dans l'administration de gènes hétérologues à une large gamme de cellules en voie de division ou non.
