Abstract:
(EN) Methods are provided for producing co-cultures of cells in which at least two types of cells are configured in a micropattern on a substrate such that modulating of the extent of heterotypic cell-cell contacts can be used to modulate metabolic and/or synthetic functions of cells. Co-culture can be carried out by coating a protein such as collagen on a substrate so that the protein defines a micropattern on the substrate, contacting the coated substrate with cells of a first type such as hepatocytes in a medium to bind the cells to the protein to produce a micropatterned cell-coated substrate, contacting the micropatterned substrate with cells of a second type such as fibroblasts in a second medium to bind the cells to the substrate. The first or second medium is a non-attachment medium such as a serum-free medium and the other medium is an attachment medium such as a medium containing serum. The attachment medium may contain an attachment factor such as fibronectin or an antibody. The micropattern may be formed by islands ofcells of either the first or second type surrounded by cells of the other type.
(FR) L'invention concerne des procédés de production de co-cultures de cellules dont au moins deux types sont configurés dans un micromodèle sur un substrat de façon que la modulation de l'étendue des contacts cellules-cellules hétérotypiques puisse être utilisée pour moduler des fonctions métaboliques et/ou synthétiques des cellules. On peut mettre en oeuvre la co-culture en revêtant une protéine telle que le collagène sur un substrat de façon que la protéine définisse un micromodèle sur le substrat, en mettant en contact le substrat revêtu avec des cellules d'un premier type telles que les hépatocytes dans un milieu afin de lier les cellules avec la protéine dans le but de produire un substrat revêtu de cellules micromodélisées et en mettant en contact le substrat micromodélisé avec les cellules d'un second type telles que les fibroblastes dans un second milieu afin de lier les cellules au substrat. Les deux milieux sont pour l'un, un milieu de non fixation, tel qu'un milieu exempt de sérum, et pour l'autre, un milieu de fixation tel qu'un milieu renfermant du sérum. Le milieu de fixation peut contenir un facteur de fixation tel que la fibronectine ou un anticorps. Le micromodèle peut être formé d'îlots de cellules du premier ou second type entourés de cellules de l'autre type.
