Abstract:
(EN) The invention features a modified small nucleolar ribonucleic acid (snoRNA) that directs the conversion of a uridine to a pseudouridine in a target nucleic acid, e.g., RNA, that includes first and second flanking regions located on either side of the uridine. The modified snoRNA includes a ribonucleotide sequence of a box H/ACA snoRNA including a Domain A sequence and a Domain B sequence, or a Domain A sequence and a Domain C sequence, wherein the ribonucleotide sequence of a box H/ACA snoRNA is modified in that the Domain A sequence is replaced by a first recognition sequence complementary to at least three consecutive nucleotides in the first flanking region in the target nucleic acid, and the Domain B or C sequence is replaced by a second recognition sequence complementary to at least three consecutive nucleotides in the second flanking region in the target nucleic acid.
(FR) L'invention a trait à un petit acide ribonucléique nucléolaire modifié (ARNsno) permettant de commander la transformation d'une uridine en une pseudouridine dans un acide nucléique cible, p. ex. de l'ARN, qui comporte deux régions flanquantes situées de chaque côté de l'uridine. L'ARNsno modifié possède une séquence de ribonucléotides d'une boîte H/ACA ARNsno comportant une séquence de domaine A et une séquence de domaine B, ou une séquence de domaine A et une séquence de domaine C, la séquence de ribonucléotides d'une boîte H/ACA ARNsno étant modifiée de sorte que la séquence de domaine A est remplacée par une première séquence de reconnaissance complémentaire à au moins trois nucléotides consécutifs de la première région flanquante dans l'acide nucléique cible, et la séquence de domaine B ou C est remplacée par une deuxième séquence de reconnaissance complémentaire à au moins trois nucléotides consécutifs de la deuxième région flanquante dans l'acide nucléique cible.
