Abstract:
(EN) Multipotent neuroepithelial stem cells and lineage-restricted oligodendrocyte-astrocyte precursor cells are described. The neuroepithelial stem cells are capable of self-renewal and of differentiation into neurons, astrocytes, and oligodendrocytes. The oligodendrocyte-astrocyte precursor cells are derived from neuroepithelial stem cells, are capable of self-renewal, and can differentiate into oligodendrocytes and astrocytes, but not neurons. Methods of generating, isolating, and culturing such neuroepithelial stem cells and oligodendrocyte-astrocyte precursor cells are also disclosed. A method of generating neural crest stem cells involves inducing neuroepithelial stem cells to differentiate in vitro into neural crest stem cells. Differentiation can be induced by replating the cells on laminin, withdrawing mitogens, or adding dorsalizing agents to the growth medium. Derivatives of the peripheral nervous system can be generated by inducing the neural crest stem cells to differentiate in vitro.
(FR) L'invention concerne des cellules souches neuro-épithéliales pluripotentes et des cellules précurseurs d'oligodendrocytes-astrocytes à lignage restreint. Les cellules souches neuro-épithéliales sont capables de s'autorenouveler et de se différencier en neurones, en astrocytes et en oligodendrocytes. Les cellules précurseurs d'oligodendrocytes-astrocytes sont obtenues à partir de cellules souches neuro-épithéliales, sont capables de s'autorenouveler et peuvent se différencier en oligodendrocytes et en astrocytes, mais pas en neurones. Des procédés permettant de produire, d'isoler et de cultiver de telles cellules souches neuro-épithéliales et cellules précurseurs d'oligodendrocytes-astrocytes sont également décrits. Un procédé permettant de produire des cellules souches de crête neurale comporte l'étape consistant à provoquer une différenciation de cellules souches neuro-épithéliales in vitro en cellules souches de crête neurale. La différenciation peut être provoquée par réensemencement des cellules sur laminine, par retrait de mitogènes, ou par ajout d'agents dorsalisants au milieu de croissance. Des dérivés du système nerveux périphérique peuvent être produits par la provocation d'une différenciation in vitro des cellules souches de crête neurale.
