Abstract:
(EN) The regulation of gene expression is of fundamental importance to all biological functions including adaptation to environmental conditions, cell division and differentiation, and the development of disease states such as cancer. The application of 'reporter' gene systems, which allow changes in gene expression to be assayed quickly and easily, has contributed greatly to our understanding of the mechanisms involved in gene regulation. A reporter gene system comprising a gene encoding an $g(a)-amylase enzyme for use in transforming mammalian cells is described. In a further example the reporter gene system comprises a second $g(a)-amylase gene, which codes for an enzyme with an electrophoretically distinct mobility from that of the first $g(a)-amylase. The two genes are used for simultaneous expression in mammalian cells, with one isozyme acting as an internal control, and the other isozyme acting as the indicator gene. The use of $g(a)-amylase as a secreted reporter gene is particularly attractive because the enzyme is extremely stable over a wide range of conditions. The measurement of $g(a)-amylase activity is simple, quantitative, sensitive, safe and inexpensive, and the range of electrophoretically distinguishable variants available allows the assays to be performed in virtually any host without interference from endogenous amylase activities.
(FR) La régulation de l'expression génétique est d'une importance fondamentale pour toutes les fonctions biologiques, notamment l'adaptation aux conditions environnementales, la division et la différentiation cellulaires et le développement d'états maladifs tels que le cancer. L'application de systèmes de gènes 'marqueurs', qui permet d'analyser rapidement et facilement des modifications de l'expression génétique, a grandement contribué à la compréhension des mécanismes impliqués dans la régulation de l'expression génétique. L'invention concerne un système de gènes marqueurs contenant un gène codant une enzyme $g(a)-amylase destinée à être utilisée dans la transformation de cellules de mammifères. Dans une autre réalisation, le système de gènes marqueurs contient un second gène d'$g(a)-amylase qui code une enzyme dont la mobilité du point de vue électrophorétique est distincte par rapport à celle de la première $g(a)-amylase.
