Abstract:
(EN) A method of collecting insect humors which permits humors to be collected from a large quantity of insects at the same time without causing the scattering of the humors, is versatile, does not cause the contamination of extracted humors with unnecessary tissues, etc., and can inhibit melanization of the collected humors. The method comprises freezing lepidopterous insects anesthetized with water, making openings in the cuticles of the frozen insects without damaging the digestive tracts, thawing the insects in a buffer solution containing a melanization inhibitor, and releasing the humors through the openings into the buffer solution by means of a spontaneous shrinkage phenomenon occurring in the process of thawing.
(FR) L'invention concerne un procédé qui permet de collecter des humeurs à partir d'une grande quantité d'insectes en même temps, sans entraîner la diffusion desdites humeurs, qui est souple, qui n'entraîne pas la contamination des humeurs extraites avec des tissus non désirés etc., et qui permet d'inhiber la mélanisation des humeurs collectés. Ledit procédé consiste à: congeler, avec de l'eau, des lépidoptères anesthésiés; pratiquer des ouvertures dans la cuticule des insectes congelés sans endommager le tube digestif; décongeler les insectes dans une solution tampon contenant un inhibiteur de mélanisation; et faire sortir les humeurs par les ouvertures de façon qu'elles pénètrent dans la solution tampon, grâce à un phénomène de retrait spontané se produisant au cours de la décongélation.
