Abstract:
(EN) There is provided a method for directly typing or sequencing HLA-A, -B, or -C alleles from a tissue sample wherein exons 2 and 3 of the HLA-A, -B, or -C alleles from the sample are amplified together in a locus specific manner and then separated out and individually amplified in a locus specific manner. After the two amplifications, the amplified exons are directly sequenced, the sequences are recombined, and a comparison is made between the derived HLA allele sequence and an HLA allele database, thereby giving an exact HLA-A, -B, or -C type for the sample being tested.
(FR) La présente invention concerne un procédé permettant de typer ou de séquencer directement les allèles HLA-A, B ou C d'un prélèvement de tissu, procédé selon lequel les exons 2 et 3 des allèles HLA-A, B ou C du prélèvement sont amplifiés ensemble de manière spécifique de locus et sont ensuite séparés et amplifiés individuellement de manière spécifique de locus. Après les deux amplifications, les exons amplifiés sont directement séquencés, les séquences sont recombinées, et une comparaison est établie entre la séquence dérivée d'allèle HLA et une base de données d'allèles HLA, ce qui permet d'obtenir un type HLA-A, B ou C exact pour le prélèvement testé.
