(EN)The present invention provides for improved methods of gene transfer, both $i(in vitro) and $i(in vivo). By treating neoplastic cells with a DNA-damaging agent prior to transduction with a transgene, the expression of the transgene is improved up to about 3-fold over the expression seen in the absence of the DNA-damaging agent treatment. This effect is not dependent on the tumor cell type, the method of DNA transduction or type of DNA-damaging agent. The effect is most dramatic when the transduction is performed about 1-3 days following treatment with the DNA-damaging agent.
(FR)Méthodes améliorées de transfert de gène, tant $i(in vitro )qu'$i(in vivo). En traitant les cellules néoplasiques au moyen d'un agent s'attaquant à l'ADN préalablement à la transduction au moyen d'un transgène, on améliore l'expression du transgène dans des proportions pouvant atteindre environ le triple des résultats obtenus en l'absence de traitement par agent s'attaquant à l'ADN. Cet effet ne dépend pas du type de cellule tumorale, de la méthode de transduction d'ADN, ni du type d'agent s'attaquant à l'ADN. L'effet est le plus évident lorsque la transduction s'effectue environ 1-3 jours à la suite du traitement par l'agent s'attaquant à l'ADN.
