(EN)A method for the production of activated tumor-specific T cells by co-cultivating, ex vivo, tumor cells from a patient with T cells from that patient, comprising the steps of: i) incubating the tumor cells with a first fusion protein obtained from a B7 protein and one partner of a biological binding pair and a second fusion protein obtained from an antibody against a cell surface antigen and the other partner of the biological binding pair; ii) inhibiting the proliferation of the tumor cells prior to or after that incubation; iii) co-cultivating the tumor cells with the T cells to be activated, until activation of the T cells is attained; iv) separating the activated T cells from the tumor cells, is highly efficient and can be carried out in a simple manner.
(FR)L'invention porte sur un procédé de production de cellules T spécifiques de tumeurs activées par co-culture ex vivo de cellules tumorales provenant d'un patient porteur de cellules T. Le procédé comporte les étapes suivantes: (i) incubation des cellules tumorales à l'aide d'une première protéine hybride obtenue à partir d'une protéine B7 et l'un des membres d'une paire biologique de liaison, et une deuxième protéine hybride obtenue à partir d'un anticorps agissant contre un antigène de surface cellulaire, et l'autre membre de la paire biologique de liaison; (ii) inhibition de la prolifération des cellules tumorales avant ou après ladite incubation; (iii) co-culture des cellules tumorales avec les cellules T à activer jusqu'à obtention de l'activation des cellules T; (iv) séparation des cellules T activées d'avec les cellules tumorales. Ledit procédé est très efficace et simple à réaliser.
