Abstract
(EN) The present invention provides a method and apparatus for rapid measurements of a fluid bulk analyte, requiring only microscale volumes. Several fluid bulk analytes can be measured simultaneously and, for biological samples, the cell content can also be measured simultaneously. The invention comprises reporter beads for chemical analysis of fluid bulk properties such as pH, oxygen saturation and ion content. Each reporter bead comprises a substrate bead having a plurality of at least one type of fluorescent reporter molecules immobilized thereon. The fluorescent properties of the reporter bead are sensitive to a corresponding analyte. Reporter beads are added to a fluid sample and the analyte concentration is determined by measuring fluorescence of individual beads, for example in a flow cytometer. Alternatively, reporter molecules which change absorbance as a function of analyte concentration can be employed.
(FR) Cette invention se rapporte à un procédé et à un appareil pour la mesure rapide d'un analyte pris dans une masse de fluide, ne nécessitant que des volumes à échelle microscopique. Plusieurs analytes pris dans une masse de fluide peuvent être mesurés simultanément et, pour des échantillons biologiques, la teneur en cellules peut également être mésurée simultanément. Cette invention concerne des perles de marquage pour l'analyse chimique des propriétés d'une masse de fluide, telles que le pH, la saturation en oxygène et la teneur en ions. Chacune de ces perles de marquage est constituée par une perle de substrat sur laquelle sont immobilisées des molécules de marquage fluorescentes d'au moins un type. Les proprétés fluorescentes des perles de marquage sont sensibles à un analyte correspondant. Des perles de marquage sont ajoutées à un échantillon fluide et la concentration de l'analyte est déterminée par mesure de la fluorescence des perles individuelles, par exemple dans un cytomètre de flux. Dans une variante, on peut utiliser des molécules de marquage qui changent d'absorbance en fonction de la concentration de l'analyte.
