Abstract

A method for assaying ammonia and/or ammonium ions contained in a liquid sample which comprises subjecting the sample to a reaction at least in the presence of an NAD synthetase, deamidated NAD, ATP, Mg2+ ions and/or Mn2+ ions, a dehydrogenase capable of forming ammonia molecules from an amine substrate and an oxidized NAD, and the amine substrate, and then determining the amounts of the ingredients consumed or generated by the reaction; and an assaying composition comprising all of the above-specified ingredients. The method is inexpensive and is effective in accurately assaying ammonia and/or ammonium ions present in minute amounts.

La présente invention concerne un procédé de dosage de l'ammoniaque, ou des ions d'ammonium, ou des deux, contenus dans un échantillon de liquide. Ce procédé comprend la mise en réaction de l'échantillon au moins en présence d'une synthétase de NAD, de NAD désamidé, d'ATP, d'ions Mg2+ ou Mn2+, ou des deux, d'une déshydrogénase capable de former des molécules d'ammoniaque à partir d'un substrat d'amine et d'un NAD oxydé, et du substrat d'amine, après quoi on détermine les quantités de produits consommées ou générées par la réaction. L'invention concerne aussi une composition de dosage comprenant tous les constituants précités. Ce procédé est peu coûteux et il est efficace pour le dosage précis de l'ammoniaque ou des ions ammonium, ou des deux, présents en faibles quantités.
