Abstract:
(EN) The present invention concerns the discovery of a new regulatory mechanism for gene expression in lactic acid bacteria (LAB), especially $i(Lactobacillus sake) LTH673 or $i(Lactobacillus plantarium) C11, that includes previously unrecognized strongly regulable promoter elements. The essential finding is that the expression of genes under the control of a promoter element dependent on the expression of the IF-K-R gene cluster. The expression of the IF-K-R gene cluster is autoinduced by the secreted peptide encoded by IF, thus providing a regulable expression system for a desired protein. The invention further comprises the purified IF protein.
(FR) La présente invention concerne la découverte d'un nouveau mécanisme régulateur destiné à l'expression de gènes dans des bactéries d'acide lactique (LAB), en particulier $i(Lactobacillus sake) LTH673 ou $i(Lactobacillus plantarium) C11, lequel comprend des éléments promoteurs pouvant être fortement régulés, lesquels n'avaient pas été reconnus auparavant. La découverte essentielle est l'expression de gènes sous le contrôle d'un élément promoteur dépendant de l'expression de la grappe de gènes IF-K-R. L'expression de la grappe de gènes IF-K-R est auto-induite par le peptide sécrété qui est encodé par IF, donnant ainsi un système d'expression régulable pour une protéine désirée. L'invention concerne en outre la protéine IF purifiée.
