Abstract:
(EN) The present invention relates to compositions and methods for inhibiting the aging of amino-containing amino acid, peptides, proteins and biomolecules. Accordingly, a composition is disclosed which comprises an agent or compound capable of reacting with the glycosyl-amino moiety of the early glycosylation product (also known as the Amadori product or the Heyns product) formed by the reaction of glucose, or other reactive sugars, with an amino-containing peptide, protein or biomolecule, thus stabilizing this early glycosylation product, and preventing its further reaction to form open-chain, carbonyl-containing advanced glycosylation end products. Suitable agents may contain a reactive aldehyde group. A preferred agent is acetaldehyde. The method comprises contacting the target biomolecule with the composition. The compounds so obtained are represented by formulae (VI) and (VIII) wherein R is the residue of an amino-containing biomolecule such as a peptide and CH2R' is the residue of the reactive aldhehyde group. Both industrial and therapeutic applications for the invention are envisioned, as food spoilage and animal protein aging can be treated.
(FR) La présente invention concerne des compositions et des procédés de prévention du vieillissement des acides aminés, peptides, protéines et biomolécules contenant un groupe amino. La composition de l'invention comprend un agent ou un composé capable de réagir avec un groupe glycosyle-amino d'un produit précoce de glycolysation (également connu comme produit d'Amadori ou produit de Heyns) obtenu par réaction du glucose ou d'autres sucres réactifs avec un peptide, une protéine ou une biomolécule contenant un groupe amino. On arrive ainsi à stabiliser le produit précoce de glycosylation et à l'empêcher de de continuer de réagir pour former des produits finis de glycosylation avancée à chaîne ouverte contenant un carbonyle. Certains agents appropriés peuvent contenir un groupe aldéhyde réactif. Un agent préféré est l'acétaldéhyde. Le procédé consiste à mettre en contact la biomolécule cible avec la composition. Les composés ainsi obtenus sont représentés par les formules générales (VI) et (VIII). Dans ces formules, R est un reste d'une biomolécule contenant un groupe amino, un peptide par exemple, et CH2R' est le reste du groupe alhéhyde de réaction. On envisage pour l'invention des applications aussi bien industrielles que thérapeutiques telles que la prévention de l'altération des aliments et le vieillissement des protéines animales.
