Abstract:
(EN) An efficient method for cloning a DNA which encodes a ligand-bound protein. The method comprises incubating on a plate a host into which a DNA inserted into an expression vector has been introduced; bringing a membrane into contact with the plate to thereby transfer the protein thus expressed from the plate onto the membrane; peeling off the membrane from the plate; adding a labeled ligand to the membrane so as to bind the ligand to the protein; detecting the label bound to the protein; and then isolating the DNA from a clone on the plate corresponding to the positive site on the membrane.
(FR) Cette invention concerne un procédé efficace de clonage d'un ADN codant une protéine liée à un ligand. Ce procédé consiste à incuber, sur une plaquette, un hôte dans lequel on a introduit un ADN qui est lui-même inséré dans un vecteur d'expression; à mettre une membrane en contact avec la plaquette de sorte que la protéine ainsi exprimée soit transférée de la plaquette à la membrane; à enlever la membrane de la plaquette; à ajouter un ligand marqué sur la membrane de sorte que le ligand se lie à la protéine; à détecter le marqueur lié à la protéine; et, enfin, à isoler l'ADN à partir d'un clone sur la plaquette correspondant au site positif de la membrane.
