Abstract:
(EN) A size exclusion membrane, particularly an immunoprotective membrane, is formed by filling the pores of a supporting membrane with a hydrogel and cross-linking the hydrogel in a hydrated state. The supporting membrane is a porous membrane having pore size of less than 20 $g(m)m and a tortuosity greater than 1 and preferably greater than 1.2. In a preferred embodiment, the supporting membrane is an open cell polyethersulfone wherein the pores are filled with a hydrated polyvinyl alcohol hydrogel which is cross-linked with, for example, glutaraldehyde while in a hydrated state. This can be used in a variety of different applications such as drug delivery, in vitro and in vivo filtration, and, for example, protection of pancreatic islet cells to provide a bio-artificial pancreas.
(FR) L'invention porte sur une membrane de filtration sur gel, une membrane immunoprotectrice notamment, constituée par remplissage des pores d'une membrane de support à l'aide d'hydrogel ensuite réticulé en état d'hydratation. La membrane de support est une membrane poreuse dont la dimension des pores est inférieure à 20 $g(m)m et dont la tortuosité est supérieure à 1 et, de préférence, à 1,2. Dans un mode de réalisation préféré, cette membrane de support est un polyéther sulfone à alvéoles ouvertes dont les pores sont remplis d'un hydrogel d'alcool polyvinylique hydraté réticulé avec de l'aldéhyde glutarique alors qu'il se trouve en état d'hydratation. Cette membrane peut être utilisée dans des applications multiples et différentes, comme l'administration de médicaments, la filtration in vitro et in vivo et, par exemple, pour la protection de cellules des îlots pancréatiques pour obtenir un pancréas bio-artificiel.
