Abstract:
(EN) The present invention is directed to a method of detecting dioxin-like compounds in a test sample. The test sample is contacted with a heteromer formed from a plurality of proteins, one of which is an active $i(Ah) receptor. If dioxin-like compounds are present in the test sample, they will bind to the $i(Ah) receptor causing it to dissociate from the heteromer as a complex containing active $i(Ah) receptor bound to a dioxin-like compound ligand. The presence of the complex is then detected. The process of the present invention can be practiced utilizing solid phase capture, competitive, and sandwich immunoassay test kit formats.
(FR) La présente invention concerne un procédé de détection de composés du type dioxine dans un échantillon test. Cet échantillon test est mis en contact avec un hétéromère se composant de plusieurs protéines, l'une d'entre elles étant un récepteur $i(Ah) inactif. Dans le cas où des composés du type dioxine sont présents dans l'échantillon test, ces derniers vont se lier au récepteur $i(Ah), ce qui va entraîner sa dissociation de l'hétéromère sous forme d'un complexe contenant le récepteur $i(Ah) actif lié à un ligand du composé du type dioxine, la présence du complexe étant ensuite détectée. Le processus décrit dans la présente invention peut être mis en oeuvre à l'aide de nécessaires de test en trousse pour des dosages immunologiques de type sandwich, compétitifs ou de capture en phase solide.
