Abstract:
(EN) Fucose and fucose analogs are synthesized enzymatically in a three step synthetic protocol. In the first step, L-fuculose-1-phosphate or analog thereof is produced by means of an aldolase catalyzed aldol addition reaction. In the second step, the fuculose-1-phosphate or analog thereof is dephosphorylated to form L-fuculose or an analog thereof using acid dephosphorylase as a catalyst. In the third step, the L-fuculose or analog thereof is converted to L-fucose or analog thereof using L-fucose isomerase as a catalyst. The synthesis may be a one pot reaction involving the addition substrates and each of the above enzymes to a single reaction vessel. Alternatively, the synthesis may be carried out with purification steps after each reaction.
(FR) On a synthétisé du fucose et des analogues de fucose par voie enzymatique dans un protocole de synthèse en trois étapes. Dans la première étape, un L-fuculose-1-phosphate ou un analogue de celui-ci est produit au moyen d'une réaction d'addition d'aldol catalysée par aldolase. Dans la seconde étape, le fuculose-1-phosphate ou son analogue est déphosphorilé pour former un L-fuculose ou un analogue de celui-ci à l'aide d'une déphosphorilase acide en tant que catalyseur. Dans la troisième étape, le L-fuculose ou son analogue est converti en L-fucose ou un analogue de celui-ci, à l'aide d'un L-fucose isomérase en tant que catalyseur. La synthèse peut être une réaction monotope faisant intervenir les substrats d'addition et chacune des enzymes précitées dans un seul récipient de réaction. La synthèse peut également faire intervenir des étapes de purification après chaque réaction.
