Abstract:
(EN) A process for producing monomeric signal chain Fv antibody molecules in insect host cells is disclosed which comprises transforming insect host cells with an expression vector comprising a DNA sequence encoding a single chain Fv antibody polypeptide and a signal peptide, under the control of which the single chain Fv antibody molecule is secreted, and subjecting the secreted single chain Fv antibody molecules to a size separation step in order to isolate monomeric single chain Fv antibody molecules from oligomers thereof.
(FR) On décrit un procédé de production de molécules d'anticorps Fv à simple chaîne monomère dans des cellules hôtes d'insecte, ce procédé consistant à transformer ces cellules hôtes à l'aide d'un vecteur d'expression comprenant une séquence d'ADN codant un polypeptide d'anticorps Fv à simple chaîne, ainsi qu'un peptide signal, sous la commande duquel la molécule d'anticorps Fv à simple chaîne est sécrétée, puis à soumettre ces molécules d'anticorps Fv à simple chaîne, sécrétées, à une étape de séparation par taille, de manière à isoler les molécules d'anticorps Fv à simple chaîne monomère à partir des oligomères de celles-ci.
