Abstract

Total bilirubin or direct bilirubin is determined by reacting a biological fluid sample with nitrous acid as an oxidizing agent and measuring optical changes in the sample. To determine direct bilirubin, the sample is reacted with nitrous acid in the presence of thiourea which suppresses the reaction of indirect bilirubin. To determine total bilirubin, the sample is reacted with nitrous acid in the presence of cetyltrimethylammonium bromide which promotes the reaction. Alternatively, the biological fluid sample is reacted with an oxidizing agent in the presence of a specified nonionic surfactant to thereby determine direct bilirubin. These determination methods well correlate to the conventional enzymatic methods. Moreover, they are highly safe methods for determining bilirubin because of being excellent in the stabilities of reagents and causing little environmental pollution.

On procède à une détermination quantitative de la bilirubine totale ou de la bilirubine directe, en faisant réagir un échantillon de fluide biologique avec de l'acide nitreux comme agent oxydant et en mesurant les changements optiques se produisant dans ledit échantillon. Pour procéder à une détermination quantitative de la bilirubine directe, on fait réagir ledit échantillon avec de l'acide nitreux en présence de thio-urée, ce qui supprime la réaction de la bilirubine indirecte. Pour procéder à une détermination quantitative de la bilirubine totale, on fait réagir ledit échantillon avec de l'acide nitreux en présence de bromure de cétyltriméthylammonium, ce qui favorise la réaction. Dans une variante, ledit échantillon de fluide biologique est mis en réaction avec un agent oxydant en présence d'un tensio-actif non ionique spécifié, pour permettre une détermination quantitative de la bilirubine directe. Ces procédés de détermination correspondent bien aux procédés enzymatiques traditionnels. Ils sont en outre hautement sûrs, car ils présentent d'excellentes stabilités des réactifs et ils ne causent guère de pollution dans l'environnement.
