Abstract

Delivery of bioactive molecules such as nucleic acid molecules encoding a protein can be significantly enhanced by immobilization of the bioactive molecule in a polymeric material adjacent to the cells where delivery is desired, where the bioactive molecule is encapsulated in a vehicle such as liposomes which facilitates transfer of the bioactive molecules into the targeted tissue. Targeting of the bioactive molecules can also be achieved by selection of an encapsulating medium of an appropriate size whereby the medium serves to deliver the molecules to a particular target. For example, encapsulation of nucleic acid molecules or biologically active proteins within biodegradable, biocompatible polymeric microparticles which are appropriately sized to infiltrate, but remain trapped within, the capillary beds and alveoli of the lungs can be used for targeted delivery to these regions of the body following administration to a patient by infusion or injection.

Il est possible d'améliorer considérablement l'administration de molécules bio-actives telles que les molécules d'acide nucléique codant pour une protéine en immobilisant la molécule bio-active dans une matière polymère avoisinant les cellules visées. La molécule bioactive est encapsulée dans un véhicule tel que des liposomes afin de faciliter le passage des molécules bioactives au tissu cible. On peut également réaliser le ciblage des molécules bioactives au tissu cible. On peut également réaliser le ciblage des molécules bio-actives en sélectionnant un support d'encapsulement de dimension adéquate de manière à ce que le support puisse acheminer les molécules vers une cible particulière. Par exemple, l'encapsulement de molécules d'acide nucléique ou de protéines bio-actives dans des microparticules polymères, biocompatibles et biodégradables, de dimension suffisante pour qu'elles puissent être filtrées par les lits capillaires et les alvéoles des poumons peut être utilisé pour réaliser une administration ciblée dans ces parties du corps après perfusion ou injection.
