Abstract

Recombinant vector carrying a gene coding for a promotor-dependent kinase and capable, when expressed in the cells transformed by the vector, of bringing out the phosphorylation of nucleoside analogues such as aciclovir or ganciclovir leading to the death of the cell by blocking the replication process. The recombinant vector is characterized in that the kinase-coding gene is the thymidine kinase gene of type 1 herpes simplex virus (HSV1-TK) deleted in the 5' portion of all or part of the sequence upstream from the second ATG initiation codon corresponding to the methionine 46 of the complete protein.

Vecteur recombinant porteur d'un gène codant pour une kinase sous la dépendance d'un promoteur et apte, quand il est exprimé dans des cellules transformées par ledit vecteur, à phosphoryler des analogues de nucléosides tels l'aciclovir ou le ganciclovir conduisant à la mort de la cellule par blocage de la réplication, caractérisé en ce que le gène codant pour une kinase est le gène de la thymidine kinase du virus Herpès simplex de type 1 (HSV1-TK) délété dans sa partie 5' de tout ou partie de la séquence en amont du deuxième codon d'initiation ATG correspondant à la méthionine 46 de la protéine complète.
