Novel N-4 modified pyrimidine analogs are provided having the structure (I) or (II), wherein: R1 is selected from the group consisting of hydrogen, acid-sensitive, base-stable blocking groups and acyl capping groups; R2 is lower alkyl; R3 is C1-C12 alkylene containing 0 to 6 linkages selected from the group consisting of -O-, -S- and -NH-; R4 is (a) in which R9 is hydrogen or an optionally substituted aliphatic group; R10 and R11 are hydrocarbyl or together form a mono- or polyheterocyclic ring; R5 is hydrogen or lower alkyl; R6 is selected from the group consisting of hydrogen, methyl, bromo and iodo; R7 is C1-C12 alkylene containing 0 to 6 linkages selected from the group consisting of -O-, -S- and -NR12- wherein R12 is hydrogen or lower alkyl; and R8 is a protecting group that can be removed and replaced by reduction. Synthetic methods for preparing the compounds are provided as well, as are methods for making and using polynucleotide probes containing the novel pyrimidine analogs.

L'invention se rapporte à de nouveaux analogues de pyrimidine modifiés en N-4 qui ont la structure (I) ou (II) dans laquelle R1 est sélectionné dans le groupe constitué par l'hydrogène, des groupes bloquants stables aux bases, sensibles aux acides et des groupes acyle coiffant; R2 représente alkyle inférieur; R3 représente alkylène en C1-C12 contenant 0 à 6 liaisons sélectionnées dans le groupe constitué par -O-, -S- et -NH-; R4 a la structure (a) dans laquelle R9 représente hydrogène ou un groupe aliphatique éventuellement substitué; R10 et R11 représentent hydrocarbyle ou forment, réunis, un noyau mono- ou polyhétérocyclique; R5 représente hydrogène ou alkyle inférieur; R6 est sélectionné dans le groupe constitué par hydrogène, méthyle, bromo et iodo; R7 représente alkylène en C1-C12 contenant 0 à 6 liaisons sélectionnées dans le groupe constitué par -O-, -S- et -NH12-, où R12 représente hydrogène ou alkyle inférieur; et R8 représente un groupe protecteur qui peut être retiré et remplacé par réduction. L'invention se rapporte également à des procédés de synthèse pour la préparation des composés ainsi qu'à des procédés de fabrication et d'utilisation des sondes polynucléotidiques contenant les nouveaux analogues de pyrimidine.
