Abstract

The invention refers to a molecular presentation system in which viral proteins are foreseen as carriers for heterologous amino acid sequences. Hereby, the viral protein is derived from small insect viruses, primarily from Flock House Virus (FHV), with a known 3-dimensional structure and amino acid sequence, whereby heterologous amino acid sequences, for exemple epitopes, are inserted inthe outwards directed loops of the viral capsid protein. Moreover, the expression of the FHV capsid protein in insect cells can produce mature virus like particles (VLP) through a recombinant baculovirus.

L'invention concerne un système de présentation moléculaire, dans lequel des protéines virales sont prévues en tant que porteurs de séquences d'aminoacides hétérologues. A cet effet, la protéine virale est dérivée de virus de petits insectes, principalement du virus du poulailler, dénommé FHV (FHV = Flock House Virus), présentant une structure tridimensionnelle et une séquence d'aminoacides connues, des séquences d'aminoacides hétérologues, par exemple des épitopes, étant insérées dans les boucles tournées vers l'extérieur de la protéine-capside virale. En outre, l'expression de la protéine-capside de FHV dans des cellules d'insectes peut produire des particules viriformes dénommées VLP (VLP = virus like particles) mûres par l'intermédiaire d'un baculovirus de recombinaison.
