Abstract

A method for determining the functional activity of activated protein C in a human plasma sample. The plasma sample is incubated with (i) exogenous reagents which activate factor V and the common pathway for blood coagulation either through factor X or by inducing the presence of thrombin in a factor V dependent manner, (ii) activated exogenous protein C; and (iii) components, such as phospholipid and calcium ions, that are necessary for efficient clotting of the plasma. The reaction is monitored to detect an end point such as the clotting time or the change in colour of a chromometric or fluorometric substrate. The end point time is compared with that obtained by a normal blood sample under the same conditions and/or with the plasma sample but without the addition of activated protein C. A decrease in clotting time as compared with normal blood is indicative of an impairment of or resistance to activated protein C.

Un procédé permet de déterminer l'activité fonctionnelle de la protéine C activée dans un échantillon de plasma humain. On fait incuber celui-ci avec: (1) des réactifs exogènes qui activent le facteur V et la voie habituelle de coagulation sanguine soit par l'intermédiaire du facteur X soit en induisant la présence de thrombine d'une façon dépendant du facteur V; (2) de la protéine C exogène activée; et (3) des composantes telles que des ions de phospholipides et de calcium, qui sont nécessaires pour une coagulation efficace du plasma. On surveille la réaction pour détecter un point final tel que la durée de coagulation ou un changement de couleur d'un substrat chromométique ou fluorimétrique. On compare la durée au point final avec celle obtenue avec un échantillon sanguin normal dans les mêmes conditions et/ou avec l'échantillon de plasma mais non additionné de protéine C activée. Une baisse de la durée de coagulation, par comparaison avec le sang normal, indique une déficience de la protéine C activée ou une résistance à son encontre.
