Abstract

The present invention relates to a monoclonal antibody which forms an immunological complex with a phosphorylated epitope of a particular subclass or form of phosphorylated tau protein without forming an immunological complex with (i) fetal tau or (ii) biopsy or autopsy derived brain material from patients having died or suffering from diseases in which NFT is not a pathological hallmark. The invention also relates to a process for diagnosing brain diseases involving monoclonal antibodies of the invention. The invention also relates to a region of the tau molecule which is specifically recognized by the monoclonal antibodies of the invention. The invention also relates to kinases or phosphorylases which specifically react with the epitope recognized by these monoclonal antibodies as well as to a method for screening compounds which interfere with the activity of these kinases and phosphorylases.

Anticorps monoclonal qui forme un complexe immunologique avec un épitope phosphorylé d'une sous-classe ou forme particulière de protéine tau phosphorylée, sans formation d'un complexe immunologique avec (i) une protéine tau foetale ou (ii) de la substance obtenue par biopsie ou autopsie provenant de patients décédés ou souffrant de maladies dans lesquelles l'enchevêtrement neurofibrillaire ne constitue pas un signe pathologique. La présente invention concerne également un procédé permettant de diagnostiquer des maladies cérébrales à l'aide des anticorps monoclonaux de la présente invention. Elle concerne en outre une région de la molécule tau qui est spécifiquement reconnue par les anticorps monoclonaux de la présente invention. Elle concerne encore des kinases ou des phosphorylases qui réagissent spécifiquement avec l'épitope reconnu par ces anticorps monoclonaux ainsi qu'un procédé de sélection de composés qui interfèrent avec l'activité de ces kinases ou phosphorylases.
