Conjugates of HA protein of influenza virus suitable for formulation as a vaccine for obtaining a strong immune response to the HA protein are formed by separating whole HA protein from the influenza virus by detergent extraction or by providing whole HA protein by recombinant procedure, treating the HA protein with hydroxylamine to form free sulfhydryl groups in the cytoplasmic domain of the protein, and cross-linking the free sulfhydryl group-containing HA protein to itself using a bis-maleimide linker or to a maleimide-modified diphtheria toxoid, tetanus toxoid or influenza NP protein or other carrier molecule. The procedure is applicable to other proteins which can be separated from a cellular material,such as a virus, and which contain thioester bonds convertible to sulfhydril groups.

Conjugués de protéine hémaglutinine (HA) du virus de la grippe utilisables dans la préparation d'un vaccin et permettant d'obtenir une forte réponse immunitaire à la protéine HA, que l'on forme en séparant la protéine HA entière du virus de la grippe par extraction au détergent ou en obtenant la protéine HA entière par des procédés de recombinaison, en traitant la protéine HA avec de l'hydroxylamine pour former des groupes sulfhydryle libres dans le domaine cytoplasmique de la protéine et en réticulant la protéine HA contenant les groupes sulfhydryle libres à elle-même à l'aide d'un liant bis-maléimide ou à un toxoïde de la diphtérie, toxoïde du tétanos ou protéine NP de la grippe modifiés par maléimide, ou à toute autre molécule porteuse. Ledit procédé est applicable à d'autres protéines qui peuvent être séparées d'un matériau cellulaire, tel qu'un virus, et qui contiennent des liaisons thioester convertibles en groupes sulfhydryle.
