(EN)An isolated protein designated p27 is disclosed. The p27 protein has an apparent molecular weight of about 27 kD, and is capable of binding to and inhibiting the activation of a cyclin E - Cdk2 complex. A nucleic acid sequence encoding p27 protein is disclosed, as well as a method for producing p27 in cultured cells. $i(in vitro) assays for discovering agents which effect the activity of p27 are also provided. Methods of diagnosing and treating hypoproliferative and hyperproliferative disorders are provided.
(FR)La présente invention concerne une protéine isolée dénommée p27. La protéine p27 est caractérisée par une masse moléculaire apparent d'environ 27 kD, et elle est capable de se lier à un complexe cycline E - Cdk2 et d'en inhiber l'activation. L'invention concerne également une séquence d'acides nucléiques codant la protéine p27 ainsi qu'un procédé de production de la p27 dans des cellules de culture. L'invention concerne en outre des dosages in vitro permettant de découvrir des agents affectant l'activité de la p27. L'invention concerne enfin des procédés de diagnostic et de traitement des troubles de l'hypoprolifération et de l'hyperprolifération.
