Abstract:
(EN) A transcription control element is provided for controlling gene expression in myogenic cells. The transcription control element comprises an isolated DNA segment having an enhancer activity in cultured cells and in non-cultured myogenic cells. The transcription control element is isolated from upstream regions of genes encoding bHLH myogenic regulatory proteins. Specifically, an enhancer element from the upstream region of human $i(myo)D and an enhancer element from the upstream region of a quail $i(qmf)1 are provided. These myoblast-specific transcription control elements are capable of significantly increasing the levels of gene expression in myogenic cells and are intended to be applied in gene therapy, using myoblast transfer and microinjection techniques, wherein myoblast-specific gene expression is desired or required.
(FR) L'invention décrit un élément de régulation de transcription servant à réguler l'expression des gènes dans des cellules myogènes. L'élément de régulation de transcription comprend un segment d'ADN isolé possédant une activité d'activateur de transcription dans des cellules cultivées et dans des cellules myogènes non cultivées. On isole l'élément de régulation de transcription des régions amont des gènes codant les protéines régulatoires myogènes de bHLH. De façon spécifique, l'invention décrit un élément activateur de transcription provenant de la région amont du $i(myo)D humain, ainsi qu'un élément activateur de transcription provenant de la région amont d'un $i(qmf)1 de cailles. Ces éléments de régulation de transcription spécifiques des myoblastes sont capables d'amplifier considérablement les niveaux d'expression génique dans des cellules myogènes et sont conçus pour s'appliquer à une thérapie génique utilisant des techniques de micro-injection et de transfert de myoblastes, où une expression génique spécifique des myoblastes est souhaitée ou nécessaire.
