The present invention relates to nucleoprotein (NP) genes of the influenza A and B viruses which were constructed from virion RNA and subsequently expressed in Spodoptera frugiperda (S19) cells using the baculovirus vector, Autographa californica nuclear polyhedrosis virus (AcNPV). The baculovirus-expressed NP antigens were reactive with monoclonal antibodies specific for either type A or B NP and with anti-NP antibodies in human serum samples. The expressed influenza NP antigens comigrated in electrophoresis with NP purified from influenza A or B virions and the recombinant NP antigens represented greater than 10 % of total protein in infected cells. Dilutions of clarified S19 cell lysates were used as antigens in a standard enzyme immunoassay format to detect serum antibody specific for influenza A or B viruses. The results showed that baculovirus-expressed NP antigens can be used in reproducible and routine assays for the serodiagnosis of influenza virus infections.

L'invention concerne des gènes de la nucléoprotéine (NP) des virus grippaux A et B qui ont été construits à partir de l'ARN du virion et ont été exprimés ensuite dans des cellules de Spodoptera frugiperda (S19) à l'aide d'un baculovirus en tant que vecteur, le virus polyhédrose nucléaire $i(Autographa californica) (AcNPV). Les antigènes NP exprimés par le baculovirus réagissaient aux anticorps monoclonaux spécifiques pour la NP type A ou B et aux anticorps anti-NP dans des échantillons de sérum humain. Les antigènes de la NP grippale exprimés ont comigré sous électrophorèse avec la NP purifiée à partir des virions grippaux A ou B, et les antigènes recombinés NP représentaient plus de 10 % de la protéine totale présente dans les cellules infectées. Des dilutions de lysates clarifiés de cellules S19 ont été utilisées comme antigènes dans un format de dosage immunologique d'enzymes standard afin de détecter l'anticorps séral spécifique au virus grippal A ou B. Les résultats ont montré que les antigènes NP exprimés par le baculovirus peuvent être utilisés dans des dosages reproductibles et de routine pour la sérodiagnostique d'infections par le virus grippal.
