Abstract:
(EN) The present invention is based, in part, on the ability to perform a stepwise series of reactions which either extend or degrade a polymer by defined units, particularly by single monomer units. These stepwise reactions are preformed on polymers that are immobilized on a solid substrate in positionally distinct locations, preferably where such locations are packed on a single substrate at very high densities. The invention provides means for simultaneous parallel sequence analysis of a large number of biological polymer macromolecules, particularly polynucleotides. The invention provides apparatus and methods that may use fluorescent labels in repetitive chemical degradative steps to determine terminal monomers on solid phase immobilized polymers. Reagents which specifically recognize terminal monomers are used to label polymers at defined positions on a solid substrate.
(FR) L'invention est basée, en partie, sur la possibilité d'effectuer une série échelonnée de réactions qui soit prolongent soit dégradent un polymère par unités déterminées, particulièrement, par simples unités monomères. On effectue ces réactions échelonnées sur des polymères immobilisés sur un substrat solide, à des emplacements distincts, de préférence, où lesdits emplacements sont condensés sur un seul substrat à des densités très élevées. L'invention décrit des moyens d'analyse séquentielle simultanée et parallèle d'un grand nombre de macromolécules polymères biologiques, en particulier, des polynucléotides. L'invention décrit un dispositif et des procédés pouvant utiliser le marquage fluorescent dans des étapes de dégradation chimique répétitives, pour déterminer les monomères terminaux des polymères immobilisés en phase solide. On utilise des réactifs reconnaissant spécifiquement les monomères terminaux, pour marquer les polymères sur les positions définies d'un substrat solide.
