Abstract:
(EN) A method for labelling the 5' end of a nucleotide synthesized on a solid support involves reacting the free 5' hydroxyl with a reactant member selected to activate the 5' terminus; reacting the activated 5' terminus with a marker molecule containing a nucleophilic group; and cleaving the labelled nucleotide from the solid phase. The invention also relates to nucleotides labelled at their 5' end by the above process, wherein the nucleophilic group of the marker moiety is covalently attached directly to the 5' carbon; and to nucleotide intermediates having activated 5' termini.
(FR) Procédé permettant de marquer le terminal 5' d'un nucléotide synthétisé sur un support solide. Dans ce procédé, on fait réagir la terminaison libre 5' hydroxyle avec un élément réactif choisi pour activer le terminal 5', on fait réagir la terminaison 5' activée avec une molécule de marqueur contenant un groupe nucléophile, et on effectue le clivage du nucléotide marqué provenant de la phase solide. Cette invention concerne également des nucléotides marqués au niveau de leur terminaison 5' à l'aide du procédé décrit ci-dessus, dans lequel le groupe nucléophile de la fraction de molécule de marqueur est lié de manière covalente directement au carbone 5'; ainsi que des produits intermédiaires de nucléotides comprenant des terminaisons 5' activées.
