Abstract:
(EN) Single chain immunotoxins directed at the human transferrin receptor using PCR based methods are described. Anti-TFR(Fv)-PE40 contains DNA for the antigen binding portion (Fv) of a monoclonal antibody directed at the human transferrin receptor fused to the DNA for a 40,000 molecular weight fragment of $i(Pseudomonas) exotoxin (PE40). In another fusion protein, DT388-anti-TFR(Fv), DNA for the antigen binding portions of the anti-TFR antibody has been fused to the DNA encoding a truncated form of Diphtheria toxin. In anti-TFR(Fv)-PE40, the single chain antibody precedes the toxin whereas in DT388-anti-TFR(Fv) the single chain antibody is at the carboxyl end of toxin. The proteins encoded by these gene fusions were expressed in $i(E. coli) and purified by conventional chromatographic techniques to near homogeneity.
(FR) Immunotoxines monocaténaires dirigées sur le récepteur de la transferrine humaine, selon des procédés fondés sur l'amplification enzymatique du génome. L'anti-TFR(Fv)-PE40 contient de l'ADN pour la partie de liaison antigénique (Fv) d'un anticorps monoclonal ayant pour cible le récepteur de la transferrine humaine fusionné à l'ADN pour un fragment de poids moléculaire de 40000 d'exotoxine de $i(Pseudomonas) (PE40). Dans une autre protéine fusionnée, la DT388-anti-TFR(Fv), l'ADN pour les parties de liaison antigénique de l'anticorps anti-TFR a été fusionné avec l'ADN codant une forme tronquée de toxine diphtérique. Dans l'anti-TFR(Fv)-PE40, l'anticorps monocaténaire précède la toxine, tandis que dans la DT388-anti-TFR(Fv), l'anticorps monocaténaire se situe à l'extrémité carboxyle de la toxine. Les protéines codées par ces fusions de gènes ont été exprimées dans $i(E. Coli) et purifiées jusqu'à l'homogénéité presque parfaite par des techniques conventionnelles de chromatographie.
