Abstract:
(EN) A gene coding for BBRP42, which is a protein secreted in an earlier stage of incubation, is cloned from a genomic DNA library of $i(Bacillus brevis) to construct an expression vector containing a signal peptide coding region and a transcription controlling region. By incubating a host carrying a recombinant plasmid containing a foreign gene joined immediately downstream of the signal peptide coding region, it is possible to allow a foreign gene product to be secreted from the host in an earlier stage of the incubation where a protease is not secreted so much, thus enabling an efficient recovery of the product.
(FR) Un gène codant pour la BBRP42, qui est une protéine sécrétée à un stade précoce d'incubation, est cloné à partir d'une banque d'ADN génomique de $i(Bacillus brevis) afin de construire un vecteur d'expression renfermant une région de codage de peptides signaux et une région de régulation de transcription. L'incubation d'un hôte portant un plasmide de recombinaison contenant un gène étranger uni immédiatement en aval de la région de codage de peptides signaux permet la sécrétion d'un produit génique étranger à partir dudit hôte à un stade précoce de l'incubation où la sécrétion deprotéase n'est pas trop importante, permettant ainsi une récupération efficace du produit.
