Abstract:
(EN) This invention provides an imaging agent which comprises a polypeptide labeled with an imageable marker, such polypeptide having an amino acid sequence substantially present in the fibrin binding domain of naturally-occuring human fibronectin and being capable of binding to fibrin. The invention further provides a method wherein the imaging agent is used for imaging a fibrin-containing substance, i.e., a thrombus or atherosclerotic plaque. Further provided are plasmids for expression of polypeptides having an amino acid sequence substantially present in the fibrin binding domain of naturally-occuring human fibronectin and being capable of binding to fibrin, hosts containing these plasmids, methods of producing the polypeptides, methods of treatment using the polypeptides, and methods of recovering, refolding and reoxidizing the polypeptides. The invention also provides for purified polypeptides substantially free of other substances of human origin which have an amino acid sequence substantially present in the fibrin binding domain of naturally-occuring human fibronectin and which are capable of binding to fibrin.
(FR) Agent d'imagerie qui comprend un polypeptide marqué à l'aide d'un marqueur décelable par l'imagerie, ledit polypeptide ayant une séquence d'acides aminés pratiquement présente dans le domaine de liaison de la fibrine de fibronectine humaine naturelle et étant capable de se lier à la fibrine. La présente invention fournit en outre un procédé selon lequel l'agent d'imagerie est utilisé pour visualiser une substance contenant de la fibrine, par ex. un thrombus ou une plaque athéroscléreuse. On décrit encore des plasmides pour l'expressivité de polypeptides ayant une séquence d'acides aminés pratiquement présente dans le domaine de liaison de la fibrine de fibronectine humaine naturelle et étant capables de se lier à la fibrine, des hôtes contenant ces plasmides, des procédés de production des polypeptides, des procédés de traitement faisant appel aux polypeptides et des procédés de récupération, de repliage et de réoxydation des polypeptides. Ladite invention décrit également des polypeptides purifiés pratiquement exempts d'autres substances d'origine humaine qui ont une séquence d'acides aminés pratiquement présente dans le domaine de liaison de la fibrine de fibronectine humaine naturelle et qui sont capables de se lier à la fibrine.
