Abstract:
(EN) A biosensor for the detection and determination of the concentration of toxins by use of enzyme inhibition. Inhibition biosensors are affected by non-specific denaturation and substrate utilisation which both result in a limited operational lifetime. These problems are mitigated by providing in an environment an enzyme which is oxidised by hydrogen peroxide the oxidised enzyme being reduced by an electron transfer agent, such as ferrocene, which is itself oxidised in the process. The electron transfer agent is capable of regeneration back to the reduced state and the extend of electron transfer regeneration gives a measure of enzyme inhibition by toxin. Electro-chemical technique allows for the generation of hydrogen peroxide from oxygen in aqueous media and the reduction of oxidised electron transfer agent. Immobilisation of the enzyme to an electrode increases efficiency whilst potentially reducing denaturation. The biosensor can be used for the environmental determination of toxins like cyanide.
(FR) Capteur biologique servant à la détection et à la détermination de la concentration de toxines à l'aide de l'inhibition enzymatique. Les capteurs biologiques inhibiteurs sont touchés par la dénaturation non spécifique et par l'utilisation de substrats, ce qui écourte leur vie utile. On peut atténuer ces problèmes à l'aide d'une enzyme qui est oxydée par peroxyde d'hydrogène, l'enzyme oxydée étant désoxydée par un agent de transfert d'électrons, tel que le ferrocène, qui est lui-même oxydé au cours de ce processus. Ledit agent est apte à se régénérer pour revenir à l'état désoxydé, et l'importance de la régénération de cet agent donne une mesure de la capacité d'inhibition enzymatique de la toxine. Une technique électro-chimique permet la génération de peroxyde à partir d'hydrogène dans des milieux aqueux, et la désoxydation de l'agent de transfert d'électrons oxydé. L'immobilisation de l'enzyme sur une électrode augmente le rendement et permet une diminution potentielle de la dénaturation. Le capteur biologique peut servir à la détermination environnementale des toxines telles que le cyanure.
