Abstract:
(EN) Methods for modulating the expression of genes are disclosed comprising selecting a portion RNA coded by the gene, said RNA portion having subportions forming a secondary structure, and contacting the RNA with oligonucleotide or oligonucleotide analog which can bind with at least one said subportions of the RNA. In accordance with the preferred embodiments, oligonucleotides or oligonucleotide analogs are designed to bind to RNA secondary structures which are of significance to the expression of the gene coding for said RNA. In accordance with a preferred embodiment, methods of treatment of human immunodeficiency virus are similarly disclosed wherein the oligonucleotides or oligonucleotide analogs are targeted at the TAR, CAR or gag-pol elements of HIV.
(FR) Procédés de modulation de l'expression des gènes consistant à sélectionner une partie de l'ARN codée par le gène, ladite partie possédant des sous-parties qui constituent une structure secondaire, et à mettre l'ARN en contact avec un oligonucléotide ou un analogue oligonucléotidique apte à se lier à au moins une desdites sous-parties de l'ARN. Selon des modes de réalisation préférés, les oligonucléotides ou les analogues oligonucléotidiques sont conçus pour se lier à des structures secondaires d'ARN significatives pour l'expression du gène qui code pour ledit ARN. Dans un mode de réalisation préféré, on a prévu des procédés de traitement du virus de l'immunodéficience humaine selon lesquels on cible les oligonucléotides ou les analogues oligonucléotidiques sur les éléments TAR, CAR ou gag-pol du VIH.
