Abstract:
(EN) A method is provided for establishing factor-dependent human B cell lines capable of secreting immunoglobulin of a desired specificity and capable of long term culturing. The method includes selecting a resting B cell having immunoglobulin of the desired specificity and culturing it in the presence of an agent capable of cross-linking its CD40 surface antigens. Long term culture requires the continued presence of the cross-linking agent. Preferably, the cross-linking agent is a monoclonal antibody specific for the CD40 antigen, presented by non-replicating mammalian cells expressing Fc$g(g)-RII.
(FR) Le procédé décrit sert à établir des lignées de lymphocytes B humains avec dépendance selon le facteur, qui sont capables de sécréter de l'immunoglobuline d'une spécificité désirée et qui sont capables d'être cultivées à long terme. Ce procédé consiste à sélectionner un lymphocyte B au repos contenant de l'immunoglobuline de la spécificité désirée et à le cultiver en présence d'un agent capable de réticulation avec ses antigènes de surface CD40. La culture à long terme nécessite la présence continuelle de l'agent de réticulation. L'agent de réticulation est de préférence constitué par un anticorps monoclonal spécifique pour l'antigène CD40, qui est présenté par des lymphocytes mammifères non réplicants exprimant le récepteur Fc$g(g)-RII.
